Walk down the dog-calming aisle and almost everything says the same word: calming. But a pheromone diffuser, a fast-acting chew and a milk-derived bioactive supplement are not doing the same thing.

So What Exactly Is Lactium® — And What Is Alpha-Casozepine?
Lactium® is derived from bovine αs1-casein, a milk protein. When that casein is enzymatically hydrolysed, it releases smaller bioactive peptide fragments. One of the key peptides associated with its calming activity is alpha-casozepine.
Alpha-casozepine was characterised in the scientific literature in 2001. Later work studied the industrial αs1-casein hydrolysate, Lactium®, and its mechanism more directly.

Why Would a Milk-Derived Peptide Affect Calm?
The nervous system relies on inhibitory signalling to keep excitation from running unchecked. One of the major inhibitory systems involves GABAA receptors.
Mechanistic research on the αs1-casein hydrolysate found that blocking the benzodiazepine binding site with flumazenil antagonised its anxiolytic-like effect in a rat model. That supports a GABAA-related explanation for how the hydrolysate can influence a calming pathway.
Evidence boundary: the receptor-pathway work is largely preclinical. The dog studies below are the more relevant evidence for canine outcomes.
Does Alpha-Casozepine Actually Work in Dogs?
